Discovery links mechanism for HIV/AIDS dementia, possibly other neurological disorders, with known cancer “checkpoint” pathway

A prominent problem in AIDS is a form of dementia that robs one’s ability to concentrate and perform normal movements. Scientists at the Burnham Institute for Medical Research (Burnham) have discovered how HIV/AIDS disrupts the normal replication of stem cells in the adult brain, preventing new nerve cells from forming. Drs. Stuart Lipton, Marcus Kaul, Shu-ichi Okamoto and their colleagues uncovered a novel molecular mechanism that inhibits stem cell proliferation and that could possibly be triggered in other neurodegenerative diseases as well. These findings were made available to medical researchers today through priority publication online by the journal Cell Stem Cell.

A normally functioning adult human brain has the ability to partially replenish or repair itself through neurogenesis, the proliferation and development of adult neural progenitor/stem cells (aNPCs) into new nerve cells. Neurogenesis can take place only within specific regions of the brain, such as the dentate gyrus of the hippocampus.

The hippocampus is the brain’s central processing unit, critical to learning and memory. aNPCs differentiate, adapt, and assimilate into existing neural circuits and mature with guidance from neurotransmitters, the chemical substances that nerve cells use to communicate with one another. The brain’s self-renewal through neurogenesis is impaired in AIDS dementia, Alzheimer’s, Huntington’s, and other neurodegenerative diseases, as evidenced by a greatly reduced number of aNPCs in brain tissue from individuals suffering from these diseases.

The Burnham team focused on the determining the effect of a protein associated with AIDS, called HIV/gp120, which plays a key role in the pathogenesis of AIDS dementia.

In initial work with cell cultures in Petri dishes, the researchers methodically ruled out the possibility that HIV/gp120 would be inducing the death of stem cells and determined instead that HIV/gp120 was acting by inhibiting stem cell proliferation. Next, they confirmed these results in a special mouse strain bred to express HIV/gp120 in its brain. This mouse model for AIDS dementia mimics several features of the disease process found in humans. They observed a significant decrease in the number of proliferating stem cells in the brains of HIV/gp120-mice compared with similar tissue from normal, wild-type mice.

HIV/gp120 is known to interact with two receptors, called chemokine receptors, which are expressed on aNPCs. The researchers discovered that the same two receptors were targeted by HIV/gp120 sourced from either mouse or human brain tissue.

In search of a mechanism behind the finding that HIV/gp120 reduced proliferation of aNPCs, the scientists studied the effect of the protein on the cell cycle. Cells undergo seasons or cycles, known as G1, S, G2, and M (for mitosis, or cell division). They found that cells exposed to HIV/gp120 got stuck in the G1 or resting phase, and that the cell cycle was arrested.

Cell cycle is studied intensively by cancer researchers who have delineated certain “checkpoint” pathways that can jam cell proliferation, one of the key behaviors of cancer. Checkpoint pathways are overcome by cancers when they fool the body’s normal machinery into producing more cancerous cells. With dementia, it turns out that the opposite is true: the Burnham team discovered that HIV/AIDS could co-opt the checkpoint pathway to prevent stem cells in the brain from dividing and multiplying.

One such checkpoint pathway is modulated by an enzyme called p38 mitogen-activated protein kinase (MAPK), whose activity is known to disrupt the cell cycle. In mature nerve cells, the Burnham team had previously shown that HIV/gp120 activates the p38 MAPK pathway to contribute to cell death. Lipton and colleagues now report that the p38 MAPK pathway is also the mechanism underlying decreased stem cell proliferation in the brain associated with HIV/AIDS. Under experimental conditions, they were able to neutralize the p38 MAPK pathway and restore stem cell proliferation.

“We show for the first time how HIV/AIDS inhibits proliferation of neural stem cells and prevents the formation of new nerve cells in the adult brain,” said Dr. Stuart Lipton, Director of Burnham’s Del E. Webb Center for Neuroscience, Aging, and Stem Cell Research.

“The fact that the mechanism of action involves the p38 MAPK enzyme is fortuitous because drugs to combat that pathway are being tested for other diseases. If they prove effective, they might also work to protect the brain. Thus, this study offers real hope for combating the bad effects of HIV/AIDS on stem cells in the brain.” Lipton went on to state, “It will be important to see if HIV/AIDS acts similarly on stem cells for other organs in the human body, as this may impact on the disease process as a whole.”

Stuart A. Lipton, M.D., Ph.D. is a practicing neurologist who has worked with HIV/AIDS dementia patients since the identification of the disease in the 1980’s. His pioneering work developing drugs to protect the brain was recognized with Germany’s Ernst Jung Prize in Medicine in 2004.

Along with Lipton, Marcus Kaul, Ph.D., and Shu-ichi Okamoto, M.D., Ph.D., are co-corresponding authors on this paper. Kaul is an Assistant Professor and Okamoto a Research Assistant Professor at Burnham.

You’d have to get as busy as a bee in order to taste all the available natural therapies out there. This one’s appealing: apitherapy, the science of honey, so-to-speak.

Could apitherapy be the perfect holistic treatment? Take a closer look.

Apitherapy is the use of bee products to promote health. Products include raw honey, royal jelly, pollen, propolis, bee wax and bee venom.

Raw honey, nutrition - and bee venom?

Aside from cooking, honey has other more unfamiliar uses; it’s an anti-bacterial and can be used for dressing wounds and burns to keep them sterile and promote healing. Nice, but most people would probably rather eat it.

Honey is made by bees from flower nectar; it contains trace amounts of many vitamins and minerals.

In terms of your health, honey’s claims seem to be proving true. Honey has significant anti-microbial properties, and is a potent anti-oxidant. Choose the darker varieties, like buckwheat honey.

Apitherapy is actually a branch of alternative medicine. It is used by apitherapists to treat a wide range of medical conditions ranging from arthritis and chronic pain to serious medical conditions like cancer and stroke.

Honey is good for you - people have been consuming it since antiquity. Although there is scant scientific evidence to support health claims, the way we see it, how could you lose with apitherapy? No matter what you choose to do with honey therapeutically, you could always just eat the stuff.

Leave the venom for the apitherapist!

EU Law Now Clearly Distinguishes Between Vitamins and Supplements that Are Natural & Those Containing Synthetic Chemicals

American Association for Health Freedom (AAHF) is pleased to announce that a stunning decision was made by the European Commission and all because of our international affiliate, Alliance for Natural Health (ANH).

In a stunning move, ANH received a letter from the European Commission (EC) signed by two heads of unit, which indicates clearly that all natural sources of vitamins and minerals, which could have been subject to a ban EU-wide, will escape the draconian EU Food Supplements Directive (FSD), and will now be regulated as foods.. This means that not only will natural sources be outside of the Directive (thereby escaping the bans); this new letter reverses the position of the EC’s legal unit given to ANH in March 2006 and upholds their interpretation of the European Court of Justice (ECJ) 2005 ruling!

In a strategic move in March 2007, the ANH made multiple submissions to the FSD Positive List. (The Positives list narrowly defines which supplement forms may be marketed in European Union nations; only forms appearing on the list can be sold.) The nutrients used for these submissions were chosen carefully to test the ECJ ruling on natural sources and include wheat germ oil as a source of full spectrum vitamin E (mixed tocopherols), palm oil as a source of vitamin E tocotrienols and palm fruits as a source of provitamin A mixed carotenoids.

The letter from Mr. Demetris Vryonides, Head of Unit “Legal Affairs” and Mr. Basil Mathioudakis, Head of Unit “Food, law, nutrition and labeling”, states: “We consider that wheat germ oil can be used in food supplements without having to be authorised and included on Annex II of the Directive. For similar reasons the dossiers on mixed carotenoids, food derived folates, [and two proprietary palm blends containing carotenoids and tocotrienols respectively] will not be processed further.”

Dr Robert Verkerk, ANH executive and scientific director, said “We are delighted to finally have this clarification from the European Commission on a point of law the ANH has been aware of since the ECJ ruling. The wider implications of this for the industry are far reaching and it effectively opens the door to functional foods and supplements containing nutrients derived from natural sources.”

Note that ANH and their leader, Dr. Robert Verkerk has steadily maintained their position in the face of huge industry criticism and never lost faith in the legal opinion the ANH obtained after the ECJ ruling even when the UK Secretary of State, the Head of Legal for the EC and most of the industry rejected the natural sources argument. ANH’s mission statement is Good Science, Good Law and this win sums that up nicely.

AAHF feels that is a remarkable turn of events and will be of great relief for patients, practitioners, and consumers and is a vindication of the position that ANH has maintained throughout. It is now time to focus on the array of other regulatory challenges facing the natural health sector. The ANH will continue to maintain scrutiny over the procedures used by the European Commission and the European Food Safety Authority in the handling of the remaining dossiers submitted earlier this year. The ANH has drawn attention to significant problems in the scientific methods being used to determine maximum permitted levels of vitamins and minerals, an issue that is subject to impending EU law.

This is a huge victory that the global health freedom community has sorely needed. AAHF is pleased to support ANH in their efforts. Funds are still desperately needed to finish paying for their extensive (and now successful) legal battle. Additional funds are needed for current projects as well. You cannot be born or exist without the innate ability to heal. Your life literally depends on it - and so does our evolution.

The whole reason for being here is to heal, and this is true of all things in your life. Healing is not limited to a skinned knee.

Garden of Healing is about embracing, and in fact, loving this process. You heal in spite of yourself, and whether you like it or not. Whether you want transformation, or think you are avoiding it, you are still always healing, and this is true, outrageously enough, in every single moment of every nanosecond of your life.

You are healing emotionally, physically, mentally, and spiritually, and by doing this, we are even healing globally. Perhaps there are other ways we transform ourselves that have not yet been discovered.

Once you heal anything, there is always the next thing waiting to be transformed. Healed is not an end state; and, we wonder what might be beyond this. Whatever is beyond total and complete health, we can only wonder, and due to the nature of ever-advancing evolution, it must be unimaginable. Funny, maybe the sky truly is the limit.

Garden of Healing is a celebration of this healing process, and especially of natural healing, self-care and health care in all its forms. We like to talk about stepped-up self-care, so that you can help yourself steer clear of the sick care industry running rampant in our times.

SELF care, HEALTH care, SICK care, URGENT care, - or DEATH…

Which will you choose?

Medical science and pharmaceuticals will always have their place; there is no disputing that. We don’t want to necessarily suggest that it’s all bad either. And, it is important to understand that you can avoid this model, to a some degree, by focusing on caring for yourself as best you can. We hope to help you.

Healing can be fun and exciting, and the rewards are immeasurable. Garden of Healing is our gift to you. We hope you visit our garden of healing, find what you need, and then go live your life. We are not counting on your repeat business.

Healed is about coming home again; it is about being all that you really are. Healing is the way to get there. COME HOME.
